Circulating nucleic acids represent promising biomarkers for non-invasive, sensitive, specific, and dynamic detection and characterization of cancers. While research in the field initially mainly focused on detection of tumor-derived mutations in patients with metastatic disease, more recent work has begun to explore applications in early-stage cancer patients and led to the development of methods for non-invasive analysis of gene expression. In this presentation, I will describe work our group has done to develop and apply novel assays for detection and analysis of circulating nucleic acids, including cell free DNA and RNA.

Learning Objectives

  • Explain why first-generation circulating tumor DNA (ctDNA) assays are potentially suitable for escalating adjuvant therapy but not for de-escalating it.
  • Describe approaches that have been developed to increase the sensitivity of ctDNA MRD assays.
  • Describe potential applications of cell free RNA liquid biopsy assays.
